The Boeing 767-300, operated by North Carolina-based charter company 21 Air, crashed shortly before 2 p.m. local time on Sunday after flying from San Juan, Puerto Rico. The aircraft also struck several vehicles on the ground and caught fire.
Three of the five injured people were in critical condition, Miami Fire Chief Raied Jadallah said. More than 200 firefighters responded to the crash, extinguishing the flames, although a fuel leak remained at the site.
Miami-Dade County Mayor Daniella Levine Cava said rescue crews reached the scene within seconds of the crash.
The cause of the crash was not immediately known. The National Transportation Safety Board will investigate the incident and plans to hold a news conference on Monday.
The crash disrupted flights at one of the busiest U.S. travel hubs during the Labor Day holiday weekend, with multiple flights delayed or cancelled.
